Does engine have to be running to do diagnostics

Hi All. have had trouble getting engine started and was wondering if this adaptor will still scan for fault codes if engine not running/started. may seem like silly question but not used one before

To run Diagnostics, car’s ECU must be powered. That could be done when key is switched to ON (or II) just before Start, all dashboard lights are illuminated then.
